  1. 神经干细胞与脑发育

  大脑新皮层,属于脑乃至整个神经系统进化上出现最晚,功能最复杂的部分,人类的大脑皮层与其他哺乳动物相比,结构上大而厚,且功能复杂。这些特点决定了人类独特的认知和创造能力。其中神经细胞数量的大量增加,是人类大脑皮层增大、厚的必要条件。我们关注神经干细胞及其亚型如何通过分裂、分化增加神经细胞的数量,从而增加脑容量。利用各种模式动物以及基因操作技术,我们将阐明大脑皮层增大、增厚的细胞与分子机制。

  2. 成体神经干细胞与微环境

  众所周知,在成年的哺乳动物的大脑中依然存在能够产生神经干细胞的区域,包括侧脑室的室管膜下区(SVZ)和海马齿状回颗粒下区(SGZ)。和胚胎期或者新生时期的发育不同,这些成体动物大脑中新生的颗粒细胞需要整合到形态和功能都已经发育成熟的微环境中(例如海马和嗅球)。而这一重塑过程不仅直接影响海马相关的学习和记忆功能,而且和许多神经精神疾病紧密相关,例如衰老引起的认知障碍、抑郁症和癫痫等。因此,系统阐明清楚成体新生神经元在整合到神经环路过程的形态和功能的微环境的因素显得尤为重要,这为利用神经干细胞的移植治疗中枢神经系统的损伤和神经退行性疾病提供新的治疗策略。

  3. 大脑皮层神经环路发育

  大脑皮层复杂的神经网络发育基础是大脑皮层发育的重要问题。神经干细胞通过分裂、分化和迁移,构成大脑新皮层的六层结构,伴随着皮层结构的形成,功能神经网络也逐渐形成。我们将利用过多光子活体时序影像技术和电生理技术,并结合生物化学,细胞生物学以及分子生物学,系统阐述大脑皮层神经网络发育的基本规律。

  4. 脑的细胞多样性与进化中的分子演化

  人类大脑皮层与低等的哺乳动物的大脑皮层相比,结构上大而厚并且有沟和回的结构。人类大脑皮层中各种细胞种类多样,包括神经元、胶质细胞等类型。仅仅就神经元来说也分为多种亚型。我们将利用单细胞测序技术,结合转基因动物等方法,分析和阐明人类大脑皮层细胞的多样性,以及这些细胞和组织结构的复杂性是如何在进化中从分子水平上进行调控的。

  5. 神经系统疾病

  神经系统疾病种类繁多,我们主要关注与大脑皮层发育、神经环路形成、以及和衰老相关的神经系统疾病。例如,头小畸形症、平滑脑症、抑郁症、孤独症、癫痫以及神经退行性疾病。目前实验室应用病人多能干细胞和已有多种疾病动物模型,对于神经系统疾病的发病机制开展细胞水平和分子水平的研究,为疾病的治疗提供理论基础和新的策略。
